TARGETING CANCER STEM CELLS WITH A CDK2 INHIBITOR IN GLIOBLASTOMA

چکیده
Abstract Glioblastoma (GB) is the most common and aggressive adult brain tumor with no cure. Brain stem cells (BTSCs) are a rare population of self-renewing multipotent in GB that contribute to tumorigenesis, therapeutic resistance, tumour recurrence. Here, we report CDK2 inhibitor suppresses BTSCs via OSM/OSMR/STAT3 signalling pathway. To begin with, performed high throughput screening (HTS) ~8400 compounds including FDA-approved drugs patient derived human naturally harbour EGFRvIII mutation elevated STAT3 phosphorylation, search can suppress EGFRvIII/OSMR/STAT3 oncogenic The screen led identification panel CDK inhibitors possessed important characteristics a) ability cross blood-brain barrier mall molecular weights 277-566 kDa, b) low Topological Polar Surface Area (TPSA) 76-115 A°, c) number Hydrogen Bond Donors (HBDs) (1-4), d) cLogP values ranging from 2-4. Following counter screens, focused on significantly efficiently reduced OSM/OSMR activation. Importantly, found compound inhibited self-renewal growth subtype BTSCs. This research has future investigation vivo assessment this combination ionizing radiation chemotherapy preclinical models GB.

Targeting Aggressive Cancer Stem Cells in Glioblastoma
Glioblastoma (GBM) is the most common and fatal type of primary brain tumor. Gliosarcoma (GSM) is a rarer and more aggressive variant of GBM that has recently been considered a potentially different disease. Current clinical treatment for both GBM and GSM includes maximal surgical resection followed by post-operative radiotherapy and concomitant and adjuvant chemotherapy. Despite recent advance...
متن کاملNatural and herbal compounds targeting breast cancer, a review based on cancer stem cells
Cancer stem cells (CSCs) are known as the major reason for therapy resistance. Recently, natural herbal compounds are suggested to have a significant role in inhibiting the breast cancer stem cells (BCSCs). The aim of this study was to explore the effective natural herbal compounds against BCSCs.This review article was designed based on the BCSCs, mechanisms of therapy resistance and natural he...
متن کاملTargeting glioblastoma cancer stem cells: the next great hope?
Glioblastoma multiforme (GBM) is the most common primary brain tumor and is notorious for its poor prognosis. The highly invasive nature of GBM and its inherent resistance to therapy lead to very high rates of recurrence. Recently, a small cohort of tumor cells, called cancer stem cells (CSCs), has been recognized as a subset of tumor cells with self-renewal ability and multilineage capacity. T...
متن کاملCancer stem cells in glioblastoma.
Tissues with defined cellular hierarchies in development and homeostasis give rise to tumors with cellular hierarchies, suggesting that tumors recapitulate specific tissues and mimic their origins. Glioblastoma (GBM) is the most prevalent and malignant primary brain tumor and contains self-renewing, tumorigenic cancer stem cells (CSCs) that contribute to tumor initiation and therapeutic resista...
متن کاملW4: Cancer Stem Cells as a New Target Point for Treatment of Glioblastoma
Glioblastoma is a destructive form of brain tumor that kills most patients within two years of diagnosis.Treatments for glioblastoma include the usual options of surgery, radiation therapy and chemotherapy, but there is no effective treatment. The tumors are capable of spreading tendrils out into the brain and it can grow back in a matter of months after being removed. The cancer stem cell...
